Abstract
Forensic DNA analysis is an enormous field that rapidly evolves and increases, which highlights a various number of disciplines such as biology, genetics, instrumentation. homicide, biophysics, statistical, recombinant DNA technology and more, that aims to resolve legal matters which revolves around rape, murder, robbery, paternity dispute or identification of human remains. A subfield of forensics is Criminalistics which includes the application of principle of branch of sciences like physical, chemical and biological sciences in civil and criminal law. Currently this subfield turned to DNA typing, DNA profiling, DNA fingerprinting and DNA analysis due to which these methods direct towards the identification of originator of a particular DNA at the crime site and offers scientifically vigorous methods for the identification of the originator of the biological material/s at the crime scene. Methods like these mostly rely on genetic polymorphisms commonly STRs (Short Tandem Repeats) which shows the highest degree of accuracy between different set of genetic information. Specificity, sensitivity and new changeless are introduced by these methods and machines which gives fewer human errors. Human errors, contamination, temperature below room temperature (24Ëš-25ËšC), low quality DNA, data analysis and misinterpretation are some challenges faced by forensic DNA analyst. This paper highlights the most crucial and important problems which DNA analyst have to deal which can compromise the validity of the result of any forensic sample and also how analysts can overcome these bottlenecks in DNA analysis. Every step and stage in DNA analysis from case receiving to sequences has problems one can face due to which they can affect the result.
